چکیده

Background Several cases of allergy to goat’s milk (GM) without allergy to cow’s milk (CM) have been reported. GM-allergy has also been reported in CM-allergic children successfully treated with oral immunotherapy. We previously demonstrated that IgE antibodies from GM-allergic/CM-tolerant patients recognize the caprine b-casein (bcap) without cross-reacting with the bovine b-casein (bbov) despite a high sequence identity (91%). We aimed in the present work to identify the critical amino acids in the non-crossreactive IgE-binding epitopes of bcap.

“Allergy to Goat/Sheep’s Milk with Good Tolerance to Cow’s Milk but Not to Cow’s Milk Cheese: Identification of 1-105 Peptide from κ-Casein as the Molecular Basis of Reactivity toward Cow’s Milk Cheese

In Europe, a new population of patients developing anaphylaxis to goat’s/sheep’s milk associated to tolerance to cow’s milk has recently appeared. Nevertheless, among these patients, some show allergic reactions to cow’s milk cheeses but these reactions are less severe than those directed against goat’s/sheep’s milk. The objective of the study was to the molecular determinants of the reactivity...

Specific IgE responses in patients allergic to goat’s milk but tolerant to cow’s milk: involvement of minor differences in primary structure between caprine and bovine caseins

Methods b-Lactoglobulin, whole casein and its four different fractions, i.e. as1-, as2-, band -caseins, were isolated from raw CM and GM. Purified b-caseins were subjected to a mild proteolysis by plasmin which generated 3 peptides, i.e. f(1-28), f(29-107) and f(108-207/9). Synthetic peptides partially recovering the N-terminal f(29107) part of the caprine b-casein were also produced. Immunorea...

Allergy to all mammalian Bovidae proteins but cow's milk in a child.

Cross reactivity between mammalian proteins (cow, goat, ewe, buffalo) has been previously described in both in vitro and in vivo studies. The highest homologies are observed between the milk proteins from cows and other Bovidae, being on average 96.1% for buffalo, 91.1% for ewe, and 87.6% for goat. Lower sequence similarities are associated with proteins contained in milks from Suidae (pigs and...

Development of β-Lactoglobulin-Specific Chimeric Human IgEκ Monoclonal Antibodies for In Vitro Safety Assessment of Whey Hydrolysates

BACKGROUND Cow's milk-derived whey hydrolysates are nutritional substitutes for allergic infants. Safety or residual allergenicity assessment of these whey hydrolysates is crucial. Currently, rat basophilic leukemia RBL-2H3 cells expressing the human IgE receptor α-chain (huFcεRIα-RBL-2H3), sensitized with serum IgE from cow's milk allergic children, are being employed to assess in vitro residu...

Application of FT near spectroscopy for determination of true protein and casein in milk

Our study deals with a possibility of determining true protein and casein in cow’s, ewe’s and goat’s milk and in ewe’s colostrums by FT NIR spectroscopy. Samples of milk were analysed by FT NIR in the reflectance mode with the transflectance cuvette. The values of correlation coefficients of calibration were as follows: cow’s protein 0.943; cow’s casein 0.964; ewe’s protein 0.997; ewe’s casein ...
